This conference will focus on the advancement of synthetic biology, especially its application in the field of antibiotic production in filamentous fungi and actinomycete bacteria, including the implementation and modification of complex biosynthesis pathway modules in existing and new production hosts. Antibiotics production is regulated by complex networks and involves intricate multi-step biosynthetic machineries, as well as major reorganization of primary metabolic fluxes to redirect cellular metabolic resources towards their biosynthesis. The urgent need for new antibiotics caused by the accelerating emergence of multi-drug resistant pathogens worldwide has led to a strong interest in the research community for decidedly novel approaches, collectively referred to as Synthetic Biology. Given the exquisitely modular nature of antibiotics biosynthesis in microorganisms and the advanced state of knowledge on it, the biotechnological manipulation of antibiotic production can be envisaged as one of the most promising first concrete industrial application domain of synthetic biology concepts. For this potential to be realized, it is critical to bring together the highly interdisciplinary community that has recently started investigating various aspects of the synthetic biology of antibiotic production. This community includes people from such diverse disciplines as metagenomics, combinatorial biochemistry, mathematical and computational modelling, cell engineering, molecular cell biology and biotechnology, and many more, both from academia and the biotechnology industry. The aim of this conference is to facilitate the indispensable mutual communication between researchers from these disciplines in a setting that stimulates the intense exchange of both fundamental knowledge and technical advancement. The direct cytopathic effects of viral replication, tissue tropism of the virus, viral-bacterial synergy, as well as innate host responses are inextricably linked and play roles to varying degrees in "seasonal," zoonotic and pandemic influenza, examples being the pandemics of 1918 and 2009 and H5N1 avian influenza. Animal models, though indispensible, have significant limitations with regard to physiological relevance to human disease. The current symposium brings together researchers working on the virus, viral receptors and tissue tropism, innate and adaptive immunity, systems biology and clinical aspects of lung injury and host defense, to address questions on the pathogenesis of influenza. The aim will be to integrate data from animal and ex vivo / in vitro human experimental models as well as human disease to understand pathogenesis of influenza and how this may lead to effective interventions. As this symposium will take place in the aftermath of the first pandemic in 40 years, there will be a wealth of new knowledge as well as intense scientific interest in the subject. These have led to unprecedented insights into structure, dynamics, and mechanisms, even in large systems, making NMR a premier tool for studying macromolecular function. Despite great progress in the field, key questions related to TGF-beta re-programming T cells are largely unknown. In this regard, it is imperative to understand the molecular mechanisms by which TGF-beta controls such a diverse T cell programming. Elucidation of these pathways would allow us to apply the knowledge and information acquired from basic studies to understand the pathogenesis of human immune-associated diseases and develop therapeutic practices in experimental and ultimately clinical settings. In addition, roles for TGF-beta in innate immunity, the mucosal immune system, autoimmunity and inflammation, anti-tumor immunity, and HIV-mediated immune deficiency have been gradually disclosed and appreciated. Thus, it is necessary to gather experts in the field together to discuss new findings and update the pivotal role of TGF-beta in the immune system, with the aim to lay out the critical and key questions and point out possible directions. A central goal is the identification of critical virus-cell crosstalk during these processes. Rather than dividing viruses into separate categories such as positive-sense RNA viruses and DNA viruses, the meeting highlights common aspects of virus lifecycles among different virus groups. The multi-disciplinary nature of the proposed meeting is important in bringing together investigators using structural, molecular, cell biological, immunological and epidemiological methods. This emphasis on shared themes and multiple experimental approaches will continue to be critical to future advances in virology. While there has been spectacular recent progress in our understanding of virus lifecycles, we are still very far from being able to design antiviral strategies, and unexpected novel aspects of virus cell biology are constantly being discovered. Plenary sessions will cover the most important aspects of virus interactions with cells. Day 1 will include cutting-edge structural virology studies and imaging methods to follow single virus particles during entry.
